Nomenclature: C57BL/6N-Tfrctm1(TFRC)Tac Slc3a2tm1(SLC3A2)Tac
The hTFRC x hCD98hc mouse is a double-humanized model designed to support in vivo evaluation of therapeutic constructs engineered to engage human transferrin receptor 1 (hTFRC/TfR1/CD71), human CD98 heavy chain (hCD98hc/SLC3A2), or both receptor targets. By placing both humanized receptors in a C57BL/6NTac background, Model 23872 supports direct comparison of alternative receptor-mediated transport strategies without introducing differences from separate host backgrounds.
The model is intended as an experimental platform for assessing construct-dependent CNS exposure, biodistribution, pharmacokinetics, target engagement, and downstream biological response when the appropriate study endpoints are included. Therapeutic performance remains dependent on construct design, receptor epitope, affinity, valency, payload, dose, route, and study conditions.
Orders by weight: Taconic cannot accept orders by weight for this model. Please note that shipments may contain animals with a larger weight variation.
Recommended Controls
The recommended control for this model is C57BL/6NTac.
Origin
Model 23872 was generated by crossing Taconic's hTFRC Mouse, Model 23236, with Taconic's hCD98hc Mouse, Model 23662, to create a double-humanized TFRC x hCD98hc line. The line has since been backcrossed for multiple generations and is congenic on the C57BL/6NTac background.

Constitutive Humanization of the Slc3a2 Extracellular Domain
The mouse Slc3a2 genomic region spanning codons V108 (exon 2) to V525 (exon 10) was replaced with the corresponding human sequence encoding the CD98hc extracellular domain. Flanking codons conserved between species (A100–P107 and A526) were retained in mouse form. Following removal of the selection cassette, the humanized allele is expressed under the control of the endogenous mouse Slc3a2 promoter, preserving the native expression pattern. One residual recombination site remains in a non-conserved region.
Constitutive Humanization of the Tfrc Extracellular Domain
The extracellular domain of mouse Tfrc was replaced with the corresponding human sequence (NCBI reference transcripts NM_011638.4, mouse; NM_001128148.1, human), producing a chimeric receptor with mouse cytoplasmic and transmembrane domains and the human extracellular domain. Following removal of the selection markers, the humanized allele is expressed under the control of the endogenous mouse Tfrc promoter, preserving the native expression pattern. Residual recombination sites remain in non-conserved regions.
