The hG-CSF KI huNOG is a HIS mouse made by engrafting hematopoietic stem cells (HSCs) in the hG-CSF KI NOG strain (no. 23225).

  • CD16+ CD89+ mature human neutrophils are elevated at 4 weeks post engraftment.
  • The study window for this model is between 1 and 8 weeks post engraftment, however, some donors may allow for longer experimental windows.
  • Human CD45 engraftment is accelerated compared to NOG or NOG-EXL strains, with the majority of animals passing 25% chimerism at 4WPE.
  • All mice are made to order and can be purchased as male or female.

Origin

The hG-CSF KI NOG mouse was developed by Ryoji Ito and colleagues at the Central Institute for Experimental Animals. To generate the model, a human granulocyte colony-stimulating factor (hG-CSF/CSF3) expression cassette driven by the CMV promoter was targeted into the murine Csf3r (G-CSF receptor) locus of the CIEA NOG mouse®. This strategy enabled systemic expression of human G-CSF while disrupting the endogenous mouse G-CSF receptor pathway, thereby preferentially supporting human neutrophil differentiation following engraftment with human hematopoietic stem cells. The colony is maintained by homozygous breeding.
