  • The FcResolv® huNOG-EXL is a HIS mouse made by engrafting human CD34+ hematopoietic stem cells (HSCs) in the FcResolv NOG-EXL strain, made to order using validated HSC donors and shipped within 1-2 weeks after engraftment
  • Post-engraftment human immune cell differentiation and QC takes place at the customer site, maximizing the potential study window.
  • Ideal for engraftment of slow growing tumors, longer treatment paradigms, or various study customizations. 
  • While engraftment failures for whole donor lots are rare, Taconic chooses donors with proven engraftment success to generate FcResolv huNOG-EXL EA mice in order to reduce the risk of lot failure. Customers should typically expect 67% of FcResolv huNOG-EXL EA mice to meet the QC standard of ≥25% hCD45+ at 10 weeks post-engraftment. Because Taconic cannot control housing and husbandry after the mice are delivered to the customer, Taconic cannot guarantee long term health or survival, chimerism, or any other study performance parameter.   • Humanized immune system mice require special housing and husbandry. FcResolv NOG-EXL Characteristics 

  • The FcResolv® NOG-EXL is a super immunodeficient NOG mouse that expresses human GM-CSF and IL-3 cytokines and has a further reduction in residual murine immune activity via ablation of mouse Fc gamma receptors (FcγRs) to reduce murine immune cell interactions with antibody-based drugs
  • This model can be used in a similar fashion to the NOG-EXL mouse, including for xenograft and immune system humanization experiments
  • Murine FcγRs can confound experiments in several ways, leading to false positive or negative results. Removing murine FcγRs provides improved accuracy for efficacy assessment of antibody-based therapies which include an Fc domain.
    • Lacks all murine FcγRs, including the FcγRI, IIB, III and IV types, along with the high affinity FcεRI receptor. The low affinity FcεRII receptor remains present
 
  • The NOG-EXL model was designed to express relatively low cytokine levels, which maintains the long-term stability of engraftment by preventing the exhaustion of the stem cell pool. This stability makes it a flexible platform for the generation of humanized mice.
    • Supports myeloid lineage engraftment through human IL-3 and GM-CSF cytokine expression
    • More efficient CD34+ human hematopoietic stem cell (HSC) engraftment
    • Higher levels of myeloid cell differentiation following human HSC engraftment
    • Low cytokine expression and successful long-term HSC engraftment
    • NOG-EXL mice are a superior host for acute myeloid leukemia (AML) patient-derived xenografts.

Lifespan: All myeloid-supportive HIS mice have limited lifespans due to a range of outcomes including anemia, thrombocytopenia, macrophage activation syndrome and hemophagocytic lymphohistiocytosis. The huNOG-EXL has the longest demonstrated lifespan compared to competing models, but this varies by donor and can be impacted by environmental and experimental factors. While Taconic has observed that huNOG-EXL mice can often survive for 25+ weeks post-engraftment (WPE), researchers should plan for studies to be complete by 23-26 WPE.

Origin

FcResolv NOG-EXL mice engrafted with human CD34+ hematopoietic stem cells (HSCs) stably develop extensive cell lineages as early as 6 to 8 weeks post-injection. Both myeloid and lymphoid lineage cells are present in peripheral blood, bone marrow, thymus and spleen and non-lymphoid tissue including lung and liver. While not all human immune cell types have been exhaustively characterized in these mice, extensive myeloid differentiation including granulocyte lineages (basophil, neutrophil and mast cells) are evident in blood and tissues. Antigen presenting cells (dendritic cells and macrophages) are also increased in frequency, both in blood and spleen.
