  • The targeting vector has been generated using 129SvEv DNA and transfected into 129SvEv ES cell line.
  • The N265S point mutation was introduced into Gabrb2 exon 8.
  • The neomycin resistance cassette, which is flanked by loxP sites, was introduced into intron 7.
  • Chimeric mice were produced and germ line transmission achieved.
  • The neomycin resistance gene has been removed in the constitutive PM mice by crossing to Cre-Deleter mice.
  • The N265S point mutation in exon 8 abolishes etomidate binding of the Gabrb2 N265S receptor, but keeps the Gabrb2 function otherwise normal.
  • Gabrb2 N265S constitutive PM mice are in a mixed C57BL/6-129SvEv genetic background.

Please note that in rare instances (2%) cryorecovery may prove to be difficult, either extending timeline or ultimately successful recovery.
