Nomenclature: C57BL/6NTac–Glp1rtm10021Tac
The model is a conditional knockout (cKO) of the Glp1r gene (glucagon-like peptide 1 receptor).
The Glp1r cKO mouse was developed by Taconic in 2026. This model was generated via conditional Knockout (KO) of the Glp1r gene. Exon 1 contains the translation initiation codon. Exon 2 was flanked by loxP sites (size of loxP-flanked region is 1.0 kb). The positive selection marker (Puromycin resistance – PuroR) was flanked by FRT sites and inserted into intron 1. The PuroR cassette also contained a tamoxifen-inducible Flpo recombinase (FlpoERT2-PuroR). The targeting vector was generated using BAC clones from the C57BL/6J RPCI-23 BAC library and was transfected into the Taconic Biosciences C57BL/6NTac ES cell line. Homologous recombinant clones were isolated using positive (PuroR) and negative (Thymidine kinase – Tk) selections. The conditional KO allele was obtained after Flp-mediated removal of the selection marker. The remaining recombination sites are located in non-conserved regions of the genome.
