• The targeting vector has been generated using C57BL/6J DNA and transfected into TaconicArtemis C57BL/6N Tac ES cell line.
• The region containing Mir34b and Mir34c have been flanked by loxP sites.
• The distal LoxP site has been inserted into a non-conserved region approx. 400 bp downstream of Mir34c. A positive selection cassette (NeoR) has been flanked by FRT recombination sites and co-inserted with the proximal LoxP site into non-conserved regions approx. 800 bp upstream of Mir34b sequence.
• The conditional KO allele has been obtained after Flp-mediated removal of the selection marker by crossing chimeras to a Flp-Deleter on a C57BL/6 background.
• The conditional KO line has been derived using C57BL/6NTac animals and the Flp-transgene was removed by segregation.
• The remaining recombination sites are located in non-conserved regions of the genome.
• The corresponding constitutive KO allele is available as line 9746.

GEM Collection models exist as cryopreserved materials. Models can typically be recovered and delivered to customers within 14-16 weeks after order receipt. Customers will receive:

• A minimum of four (4) mice mutant for the selected mutation. In rare cases where the cryopreserved material also contains a recombinase gene, this may be reduced to 1-2 mutant mice.
• Validated genotyping protocols
• Animal health report. Mice will meet Taconic's Murine Pathogen Free™ (MPF™) health standard.

Please note that in rare instances (2%) cryorecovery may prove to be difficult, either extending timeline or ultimately successful recovery.
