  • The targeting vector has been transfected into TaconicArtemis C57Bl/6 ES cell line.
  • The following elements have been inserted into the ROSA26 locus: a CAG promoter cassette; a loxP-flanked transcription termination cassette (STOP) containing a combination of polyadenylation signals; the miR30-miR122a transgene and a polyadenylation site (hGH polyA).
  • The conditional TTG line has been generated by crossing chimeras to C57BL/6NTac animals.
  • The constitutive TTG allele can be generated by Cre-mediated recombination by crossing the line to a Cre-Deleter.
  • Deletion of the STOP cassette should result in transcription of the mir122a transgene.
  • The presence of the NeoR cassette will terminate the transcription from the ROSA26 promoter.

GEM Collection models exist as cryopreserved materials. Models can typically be recovered and delivered to customers within 14-16 weeks after order receipt. Customers will receive:

  • A minimum of four (4) mice heterozygous for the selected mutation. In rare cases where the cryopreserved material also contains a recombinase gene, this may be reduced to 1-2 heterozygous mice.
  • Validated genotyping protocols
  • Animal health report. Mice will meet Taconic's Murine Pathogen Free™ (MPF™) health standard.

Please note that in rare instances (2%) cryorecovery may prove to be difficult, either extending timeline or ultimately successful recovery.
For-profit users: Commercial users may purchase available GEM Collection models by signing the GEM Collection Commercial Terms of Sale and providing a PO. The GEM Collection Commercial Terms of Sale grant perpetual use rights.
Non-profit users: Non-profit users must provide a scientific workplan and execute an MTA prior to accessing a GEM Collection Model.

Non-profit order process is as follows:

  1. Non-profit Investigator visits the Taconic website and identifies GEM of interest.
  2. Non-profit Investigator inquires on Taconic website.
  3. Taconic will answer questions about the GEM lines and the process.
  4. Taconic sends request for MTA.
  5. Non-profit Investigator provides scientific workplan answering 4 basic questions.
  6. MTA is issued for non-profit Investigator/institution review and signature.
  7. MTA is executed by all parties and release of model is approved.
  8. Taconic contacts requestor to obtain purchase order (PO) to cover the cryorecovery and shipment costs.

Taconic begins cryorecovery upon receipt of PO and delivers model to requestor. It is the requestor's obligation to determine that the GEM meets their research needs.

Access to this model for non-profit customers requires approval of a third-party Material Transfer Agreement (MTA). The terms of this MTA are non-negotiable; an example agreement is available so that you may preview the terms. MTA approval may require 4-6 months due to circumstances outside our control. Once approved and the purchase confirmed, order fulfillment will require an additional 14-16 weeks.
