  • The targeting vector has been generated using C57BL/6J DNA and transfected into TaconicArtemis C57BL/6NTac ES cell line.
  • Exon 1 contains the translation initiation codon.
  • Exons 2 to 7 have been flanked by loxP sites (size of loxP-flanked region: 2.7 kb).
  • The conditional KO allele has been generated after Flp-mediated recombination by crossing chimeras to a Flp-Deleter on a C57BL/6 background.
  • The conditional KO line has been derived using C57BL/6NTac animals and the Flp-transgene was removed by segregation.
  • The constitutive KO allele can be generated by Cre-mediated recombination. Deletion of exons 2 to 7 should result in loss of function of the Lpcat4 gene by deleting both transmembrane domains and part of the catalytic domain and by generating a frameshift from exon 1 to exon 8 (premature Stop codon in exon 9).
  • The remaining recombination sites are located in non-conserved regions of the genome.
  • According to NCBI and Ensembl databases, the Nut gene (Entrez ID: 213765; ENSMUSG00000041358) is located approx. 4.5 kb downstream of Lpcat4 exon 7 in a tail-to-tail orientation.
  • The corresponding constitutive KO allele is available as line 10815.

GEM Collection models exist as cryopreserved materials. Models can typically be recovered and delivered to customers within 14-16 weeks after order receipt. Customers will receive:

  • A minimum of four (4) mice heterozygous for the selected mutation. In rare cases where the cryopreserved material also contains a recombinase gene, this may be reduced to 1-2 heterozygous mice.
  • Validated genotyping protocols
  • Animal health report. Mice will meet Taconic's Murine Pathogen Free™ (MPF™) health standard.

Please note that in rare instances (2%) cryorecovery may prove to be difficult, either extending timeline or ultimately successful recovery.

Nonprofit users: Nonprofit users must execute an MTA prior to accessing a GEM Collection Model.
Nonprofit order process is as follows:
  1. Nonprofit Investigator visits the Taconic website and identifies GEM of interest.
  2. Nonprofit Investigator fills out MTA (including scientific workplan answering 4 basic questions) and Investigator's institution reviews, signs and sends to info@taconic.com.
  3. MTA is executed by all parties and release of model is approved.
  4. Taconic contacts requestor to obtain purchase order (PO) to cover the cryorecovery and shipment costs if this has not already been provided.

Taconic begins cryorecovery upon receipt of PO and delivers model to requestor. It is the requestor's obligation to determine that the GEM meets their research needs.

Access to this model for nonprofit customers requires approval of a third-party Material Transfer Agreement (MTA). The terms of this MTA are non-negotiable. MTA approval may require 4-6 months due to circumstances outside our control. Once approved and the purchase confirmed, order fulfillment will require an additional 14-16 weeks.
