The targeting vector has been generated using C57BL/6 DNA and transfected into C57BL/6 ES cell line.
Hcst exons 3 and 4 were removed by the neomycin resistance cassette, which is flanked by loxP sites.
The neomycin resistance gene has been deleted by transient transfection of Cre in targeted ES cells.
Chimeric mice were produced and germ line transmission achieved.
Deletion of exons 3 and 4 should result in loss of function of the Hcst gene by deleting critical domains for the expression of a functional protein.
The remaining recombination sites are located in intron regions of the genome.
