The targeting vector has been generated using 129SvEv DNA and transfected into 129SvEv ES cell line.
Gabrb2 exons 6 and 7 were deleted by the neomycin resistance cassette, which is flanked by loxP sites.
Chimeric mice were produced and germ line transmission achieved.
The neomycin resistance gene has not been deleted in the constitutive KO mice.
Deletion of exons 6 and 7 should result in loss of function of the Gabrb2 gene by deleting functional domains of the protein.
Gabrb2 constitutive KO mice are in a mixed C57BL/6-129SvEv genetic background.
