The H105R point mutation was introduced into Gabra5 exon 4.
The neomycin resistance cassette, which is flanked by loxP sites, was introduced into intron 3.
Chimeric mice were produced and germ line transmission achieved.
The neomycin resistance gene has been removed in the constitutive PM mice by crossing to Cre-Deleter mice.
The H105R point mutation in exon 4 abolishes benzodiazepine binding of the Gabra5 H105R receptor, but keeps the Gabra5 function otherwise normal.
Gabra5 H105R constitutive PM mice are in a mixed C57BL/6-129SvEv genetic background.
