The targeting vector has been generated using C57BL/6J DNA and transfected into TaconicArtemis C57BL/6NTac ES cell line.
Exon 2 contains the translation initiation codon.
Exon 2 has been flanked by loxP sites (size of loxP-flanked region: 3.0 kb).
The conditional KO allele has been generated after Flp-mediated recombination by crossing chimeras to a Flp-Deleter on a C57BL/6 background.
The conditional KO line has been derived using C57BL/6NTac animals and the Flp-transgene was removed by segregation.
The constitutive KO allele can be generated by Cre-mediated recombination. Deletion of exon 2 should result in loss of function of the Fut1 gene by deleting the complete open reading frame. After deletion of exon 2, the transcriptional STOP cassette (human Growth Hormone - pA) should prevent read-through to downstream gene (Izumo1).
The remaining recombination sites are located in non-conserved regions of the genome.
According to NCBI and Ensembl databases, there are 3 genes in the neighborhood of the Fut1 gene:
- The Fgf21 gene (Entrez ID: 56636; ENSMUSG00000030827) is located 3.1 kb of upstream of Fut1 exon 2 in a head-to-head orientation.
- The Izumo1 gene (Entrez ID: 73456; ENSMUSG00000064158) is located 751 bp downstream of Fut1 exon 2 in a tail-to-head orientation.
- The Rasip1 gene (Entrez ID: 69903; ENSMUSG00000044562) is located 6.5 kb downstream of Fut1 exon 2 in a tail-to-head orientation.
The corresponding constitutive KO allele is available as line 11297.
