  • The targeting vector has been generated using 129Sv DNA and transfected into 129Sv ES cell line.
  • Tyrobp exons 3 and 4 were removed by the neomycin resistance cassette, which is flanked by loxP sites.
  • The neomycin resistance gene has been deleted by transient transfection of Cre in targeted ES cells.
  • Chimeric mice were produced and germline transmission achieved.
  • Deletion of exons 3 and 4 should result in loss of function of the Tyrobp gene by deleting the transmembrane region and part of the cytoplasmic region.
  • The remaining recombination sites are located in intron regions of the genome.
