129S6 Background


The Rag2 mouse was developed in the laboratory of Frederick W. Alt at Columbia University. The model was created by targeting the Rag2 gene in CCE ES cells and injecting the targeted cells into blastocysts. Taconic received stock from the Alt lab in April 1992 on an inbred 129S6 background. Mice were caesarean derived in August 1992. The background differed from a 129S6 background at two biochemical markers, Gpi1a and Pgm1a. In 2000, the mice were backcrossed three generations (N3) to 129S6 to select for the Gpi1c and Pgm1b markers. The colony was maintained through matings of homozygous mice.
