Constitutive with Option for
Conditional Knock Out

This package allows the flexibility to generate both the Constitutive Knock Out and the Conditional Knock Out from the same Gene Targeting event, saving time and resources. We have developed a range of multi-purpose vectors that allow the generation of a constitutive KO mouse and then subsequently a conditional mouse without the necessity of re-making the vector or re-targeting the ES cells or generating new chimeras. All lines can be generated directly on an inbred C57BL/6 background. We routinely delete the selection marker to avoid any potential hypomorphic effects.

Generation of Constitutive Knock Out with Option for Conditional Knock Out consists of the following steps:

1. Targeting Vector Design & Construction
2. Targeted C57BL/6 ES cells
3. Generation of heterozygous mice for the constitutive knock out by crossbreeding chimeric mice with Cre deleter

Expected timeline for generation of constitutive knock out: 40 WEEKS

Upon request, option for Conditional Knock Out Mouse Model (to be exercised within 6 months of delivery of mice from above):

4. Generation of heterozygous mice for the conditional knock out by crossbreeding chimeric mice with Flp deleter

Expected timeline for generation of Conditional knock out: 12 WEEKS

Taconic has incorporated rigorous quality control and documentation of different mouse production steps using project management software and the Laboratory Information Management System (LIMS). A designated Taconic scientific project manager will ensure optimal communication and strict adherence to the timeline and deliverables throughout the project. Taconic will provide a detailed Milestone Documentation after each of the steps mentioned above.

Upon generation of the line at Taconic, we will coordinate the delivery and further breeding of your animals to produce the research cohorts you want at the health status you need.
