The targeting vector has been generated using 129Sv DNA and transfected into 129Sv ES cell line.
Tyrobp exons 3 and 4 were removed by the neomycin resistance cassette, which is flanked by loxP sites.
The neomycin resistance gene has been deleted by transient transfection of Cre in targeted ES cells.
Chimeric mice were produced and germline transmission achieved.
Deletion of exons 3 and 4 should result in loss of function of the Tyrobp gene by deleting the transmembrane region and part of the cytoplasmic region.
The remaining recombination sites are located in intron regions of the genome.
