  • The targeting vector has been generated using C57BL/6J DNA and transfected into TaconicArtemis C57BL/6N Tac ES cell line.
  • Grik1 exon 1 contains the translation initiation site.
  • Exon 4 has been flanked by LoxP sites.
  • The conditional KO allele has been obtained after Flp-mediated removal of the selection marker by crossing chimeras to a Flp-Deleter on a C57BL/6 background.
  • The conditional KO line has been derived using C57BL/6NTac animals and the Flp-transgene was removed by segregation.
  • Deletion of exon 4 (1.0 kb) should result in loss of function of the Grik1 gene by generating a frameshift from exon 3 to exons 5 to 12 (premature Stop codon in exon 5).
  • Remaining recombination sites are located in non-conserved regions of the genome.

NOTICE: Access to this model is conditioned on the requestor's institution agreeing to the terms and conditions of a Material Transfer Agreement (MTA).

Steps to initiate and place order:

  1. Nonprofit Investigator visits the Taconic website and identifies GEM of interest.
  2. Nonprofit Investigator inquires on Taconic website.
  3. Taconic will answer questions about the GEM lines and the process.
  4. Taconic sends request for MTA.
  5. Nonprofit Investigator provides scientific workplan answering 4 basic questions.
  6. MTA is issued for Nonprofit Investigator/institution review and signature.
  7. MTA is executed by all parties and release of GEM is approved.
  8. Taconic contacts requestor to obtain purchase order (PO) to cover the cryorecovery ($4,900 per cryopreserved GEM line) and shipment costs.
  9. Taconic begins cryorecovery upon receipt of PO and delivers GEM line to requestor (See pricing and Deliverables).

It is the requestor's obligation to determine that the GEM meets their research needs.

Upon execution of the MTA terms of use, requestor needs to provide purchase order (PO) to cover the cryorecovery ($4,900 per cryopreserved GEM line) and additional shipment costs.

  1. Taconic will begin standard derivation of cryopreserved material (sperm or embryos) upon receipt of PO from recipient.
  2. Taconic will conduct testing using its International Health Monitoring System™ (IHMS™) to ensure that the mice meet a Murine Pathogen Free™ (MPF™) health status.
  3. Taconic will genotype the recovered mice prior to shipment.
  4. Taconic will provide a health status report, the genotyping results of the recovered animals and genotyping assay information for the requested GEM line.
  5. Taconic shall deliver a minimum of two (2) mice heterozygous at the selected gene of interest of a requested GEM line.
  6. Taconic will make shipping arrangements for the requested GEM to the Licensed Nonprofit recipient.
  7. Nonprofit investigator is responsible for covering the shipment costs.

Please note that in rare instances (2%) cryorecovery may prove to be difficult, either extending timeline or ultimately successful recovery.
